Is there an upgrade from iSilo 2.x to 4 or would I have to purchase version 4?

Fill out and submit the Lost Registration Code (http://www.isilo.com/support/LostRegCodeRequest.php) form to get your 3.x registration code, which you can then use to purchase the upgrade to version 4.x.
